作者用九个GMT(一种家族性周围神经病)患者的活检皮肤组织进行培养研究,从一患者的组织培养中分离到一株巨细胞病毒(GMV)。为了解该病毒的特征,作者从以下3个方面进行了初步鉴定。 1) 宿主范围实验:分别用恒河猴肾细胞系,鼠胚组织细胞,Wi—38,Detroit 550细胞,兔肾细胞,新生儿包皮细胞以及一些成人皮肤组织细胞进行接种培养,发现该病毒仅在人纤维母细胞中繁殖。 2) 电子显微镜观察:电镜检查揭示在感染细胞的胞浆和胞核内大量存在有囊膜
The authors studied the culture of biopsied skin tissue from nine patients with GMT, a familial peripheral neuropathy, and isolated a cytomegalovirus (GMV) from a patient’s tissue culture. In order to understand the characteristics of the virus, the author conducted the preliminary identification from the following three aspects. 1) Host-range experiments: Inoculation of rhesus monkey kidney cell lines, mouse embryonic tissue cells, Wi-38, Detroit 550 cells, rabbit kidney cells, neonatal foreskin cells, and adult human adult skin tissue cells were performed.
