目的:观察犬心室扩张和逆转时心肌胶原含量,Ⅰ/Ⅲ型比值变化。方法:17只家犬,随机分为(1)持续起搏(PCn=6)组,以250次/分频率起搏4周;(2)恢复(RC,n=6)组,持续右空快速起搏4周停止起搏,恢复4周;(3)对照(SO,n=5)组。PC、RC组分别于起搏和恢复4周后,SO组于假手术1周后,以羟脯氨酸法测定心肌胶原含量、十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)法测定心肌胶原Ⅰ/Ⅲ型比值。结果:与SO组比,PC组心肌胶原含量增加,Ⅰ/Ⅲ型胶原比值显著降低(P<0.05),而RC组心肌间质胶原及Ⅰ/Ⅲ型胶原比值也有相应的恢复。结论:右室快速起搏时,心肌间质胶原含量增加,Ⅰ/Ⅲ型胶原比值降低,停止起搏扩张心室逆转后,胶原合量和成分也随之恢复。
Objective: To observe the myocardial collagen content and type Ⅰ / Ⅲ ratio of ventricular dilatation and reversal in dogs. METHODS: Seventeen dogs were randomly divided into (1) continuous pacing (PCn = 6) group and pacing at 250 beats / min for 4 weeks; (2) recovery (RC, n = 6) Pacing for 4 weeks after fast pacing was stopped and recovered for 4 weeks; (3) control group (SO, n = 5). After 4 weeks of pacing and recovery respectively in PC and RC groups, the collagen content in myocardium of SO group was measured by hydroxyproline one week after sham operation, and the content of myocardial collagen was determined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS- PAGE) was used to determine the type I / III ratio of cardiac collagen. Results: Compared with SO group, the content of collagen in myocardium increased, the ratio of type Ⅰ / Ⅲ collagen decreased significantly (P <0.05), and the ratio of collagen type Ⅰ / Ⅲ in RC group also recovered correspondingly. Conclusions: During right ventricular rapid pacing, the content of collagen in myocardium increases and the ratio of type Ⅰ / Ⅲ collagen decreases. After stopping the pacing and dilating ventricular reversal, the amount and composition of collagen recovered.
