为进一步探讨丝裂霉素膀胱内灌注治疗膀胱肿瘤的机制,研究了丝裂霉素诱导人膀胱癌 B I U87 细胞凋亡的规律。形态学观察具有典型凋亡征象,胞浆浓缩,染色质凝集,核固缩、裂解及凋亡小体。凋亡指数与诱导时间呈正相关( P< 0.01);流式细胞仪 D N A 直方图上出现特征性的亚二倍体(亚 G1 )峰;凋亡过程中伴 Bcl2 基因的下调,且与诱导时间呈负相关 ( P< 001)。提示丝裂霉素诱导癌细胞凋亡是腔内化疗的重要机制,降低 Bcl2 的表达可提高膀胱癌腔内化疗的疗效。
To further explore the mechanism of intravesical instillation of mitomycin in bladder cancer, mitochondria induced apoptosis in human bladder cancer B U87 cells. Morphological observation showed typical signs of apoptosis, cytoplasm condensation, chromatin condensation, nuclear pyknosis, lysis and apoptotic bodies. The apoptotic index was positively correlated with the induction time (P <0.01), and the characteristic sub-diploid (sub G1) peak appeared on the histogram of D N A by flow cytometry. The apoptotic index Down-regulated, and was negatively correlated with induction time (P <001). Tip Mitomycin-induced apoptosis of cancer cells is an important mechanism of intra-luminal chemotherapy, reducing the expression of Bcl 2 can improve the efficacy of intra-articular chemotherapy of bladder cancer.
