目的观察骨髓间充质干细胞(BMSC)的电生理特性,为临床干细胞移植的安全性提供参考。方法分离并培养BMSC,应用全细胞膜片钳技术记录BMSC的电生理特性。结果BMSC的静息膜电位为(-33.6±6.8)mV,平均细胞膜电容为(32.3±8.7)pF(n=30)。电压钳实验显示,BMSC上可记录到KCaI、to,IKSUS和IK14种通道电流,但未记录到INa和ICa通道电流。结论BMSC上存在4种不同类型钾离子通道。
Objective To observe the electrophysiological characteristics of bone marrow mesenchymal stem cells (BMSCs) and provide a reference for the safety of clinical stem cell transplantation. Methods BMSCs were isolated and cultured. Whole-cell patch-clamp technique was used to record the electrophysiological properties of BMSCs. Results The resting membrane potential of BMSC was (-33.6 ± 6.8) mV and the average cell membrane capacitance was (32.3 ± 8.7) pF (n = 30). Voltage clamp experiments showed that KCaI, to, IKSUS and IK14 channel currents were recorded on BMSC, but no channel currents were recorded on INa and ICa channels. Conclusion There are 4 different types of potassium channels on BMSC.
