目的 :观察葡萄球菌肠毒素 A的细胞增殖诱导能力及其抗肿瘤作用。方法 :MTT微量酶反应比色法。结果 :纯化 SEA在 1 0 - 12 ~ 1 0 - 5g/ml浓度范围对体外培养的 BALB/C鼠脾细胞表现了细胞增殖诱导能力 ,并呈剂量依赖关系 ,其中 1 0 - 7~ 1 0 - 5g/ml SEA的作用强于最适量 (2 5μg/ml) PHA。在 E/T为 5~ 2 0∶ 1条件下 ,1 0 - 5g/ml SEA活化 48小时的 BALB/C鼠脾细胞对 Yac- 1细胞的杀伤活性高于 NK细胞 ,但 SEA未能增强 BALB/C鼠脾细胞对 B16细胞的杀伤活性。结论 :葡萄球菌肠毒素 A具有较强的细胞增殖诱导能力 ,据此超抗原特性可应用于肿瘤的生物治疗
Objective: To observe the cell proliferation-inducing ability of staphylococcal enterotoxin A and its anti-tumor effect. Methods: MTT micro-enzyme reaction colorimetric method. Results: Purified SEA showed the ability of inducing cell proliferation in splenocytes of BALB / c mice cultured in vitro in a concentration range of 10 ~ 12 ~ 10 ~ 5g / ml in a dose - dependent manner. Under the conditions of E / T of 5 ~ 20: 1, the killing activity of BALB / c mice spleen cells with 100-5 g / ml SEA for 48 hours on Yac-1 cells was higher than that of NK cells, but SEA failed to enhance BALB / C murine splenocytes on B16 cells. Conclusion: Staphylococcal enterotoxin A has a strong ability of inducing cell proliferation, so the characteristics of superantigen can be applied to tumor biotherapy
