We have reported that when acupuncture analgesia in rabbits, the 5-hydroxytryptamine (5-HT) content in the thalamus and medulla increased significantly, and 5-HT in the ventricle could increase the pain threshold, suggesting that acupuncture analgesia may be the same. It is related to the release of neurotransmitters such as 5-HT in the brainstem reticular formation. According to recent studies, both the dorsal raphe nucleus raphe magnus and the large nucleus raphe magnus (the site where 5-HT neurons are concentrated) have analgesic effects. If the activity of 5-HT neurons in the brain is inhibited, the effect of acupuncture analgesia is weakened. On the contrary, electrical stimulation of the nuclei (the site of concentration of adrenergic neurons in the brain stem) has the effect of inhibiting acupuncture analgesia. Destroying the nuclear spot can significantly increase the analgesic effect of acupuncture
