目的 研究胰头癌组织与正常胰腺组织中γ 谷氨酰转肽酶 (γ glutamyltranspeptidase,GGT)基因的表达及核苷酸序列的差异 ,以阐明胰头癌患者血清GGT异常升高的分子机制。方法 收集 3例胰头癌、2例胆管癌和 2例正常胰腺的手术切除标本。应用逆转录多聚酶链反应及核苷酸序列分析方法 ,检测不同组织中GGTmRNA表达量的变化及GGT基因核苷酸序列。结果 胰头癌与正常胰腺组织中的GGTcDNA核苷酸序列中 12 0 2bp~ 16 0 5bp相一致。在正常胰腺、胆管癌和胰头癌组织中均可见GGTmRNA的表达扩增产物 ,但胰头癌组织中GGT基因表达水平明显高于胆管癌和正常胰腺组织。结论 胰头癌与正常胰腺组织中的GGT基因序列是相同的 ,但胰头癌组织中GGTmRNA表达量明显增高 ,证实GGT是诊断胰头癌的有效指标。
Objective To study the difference of GGT gene expression and nucleotide sequence between pancreatic cancer tissues and normal pancreatic tissues so as to clarify the molecular mechanism of abnormal GGT in patients with pancreatic head cancer. Methods Three cases of pancreatic cancer, two cases of cholangiocarcinoma and two cases of normal pancreas were collected. Reverse transcription polymerase chain reaction and nucleotide sequence analysis were used to detect the changes of GGT mRNA expression in different tissues and the nucleotide sequence of GGT gene. Results The pancreatic head carcinoma was consistent with the nucleotide sequence of 12 0 2 bp to 16 0 5 bp in the GGT cDNA of normal pancreas. In normal pancreas, cholangiocarcinoma and pancreatic head cancer tissues, GGT mRNA expression was observed, but the expression of GGT gene in pancreatic head cancer was significantly higher than that in cholangiocarcinoma and normal pancreatic tissue. Conclusion The sequence of GGT gene in pancreatic cancer and normal pancreatic tissue is the same, but the expression of GGTmRNA in pancreatic cancer tissue is significantly increased, which confirms that GGT is an effective indicator for the diagnosis of pancreatic cancer.
