利用龙眼茎尖离体培养技术进行脱毒和快繁无病(鬼帚病)苗木,是目前发展龙眼生产行之有效的方法。周丽侬等以无菌实生苗和自然环境生长的刚抽出3—5厘米长的嫩枝为外植体,通过诱导愈伤组织或诱芽来繁殖试管苗,并进行了生根试验。但外植体过大,不利于外植体消毒,不能达到脱毒的目的。我们从1988年下半年开始,探索龙眼茎尖培养和脱除龙眼鬼帚病毒的可能性。经过2年多的试验,茎尖培养直接成苗获得成功,为今后进一步利用茎尖培养进行脱毒和培育无病苗提供了可能。现将研究结果初报如下:
The use of longan stem tip in vitro culture technology for virus-free and fast propagation disease-free (ghost broom disease) seedlings, is the development of longan production line effective method. Zhou Lonong and other sterile germs and natural environment just grow 3-5 cm long shoots as explants, by inducing callus or buds to reproduce the plantlets and rooting test. However, the explant is too large, not conducive to explant disinfection, can not achieve the purpose of virus-free. We started in the second half of 1988 to explore the possibility of cultivating longan shoot tips and removing longan ghost broom virus. After more than two years’ experiment, the direct success of the shoot tip culture into seedlings has made it possible to further utilize the shoot tip culture to detoxify and cultivate the disease-free seedlings in the future.
