本文报道直接用组织培养细胞从急性甲型肝炎病人的粪便提取物中分离甲型肝炎病毒(HAV)。所用组织培养细胞为恒河猴胚肾细胞(FRhK_6)。急性肝炎病人粪便标本经高速离心和过滤处理后感染单层细胞,35℃作用4小时,然后加维持液置35℃培养。在培养传代过程中,采用直接免疫荧光法(IF)检测HAV增殖情况,并对培养物进行放射免疫试验(RIA),免疫粘附血凝试验(IAHA)测定其固定补体以及引起人“O”型红细胞凝集能力,以检测HAV抗原,从而证实分离物是否为HAV。同时按常规方法进行细胞培养提取液的免疫电镜检查。此外,用1 ml 1:
This paper reports the isolation of Hepatitis A virus (HAV) from the faeces extract of patients with acute hepatitis A using tissue culture cells directly. Tissue culture cells used were rhesus embryo kidney cells (FRhK_6).
