测定味精中谷氨酸含量,多采用旋光法和中和法。由于氨基酸在碱性条件下能定量地从磷酸铜沉淀中络合铜,所以可通过原子吸收光谱法测定铜间接测定味精中谷氨酸含量。所拟方法简便,重现性好,可检出0.10mg谷氨酸,变异系数为2.62~4.71%,回收率在92~109%。用中和法与本法同时测定一些味精样品中谷氨酸含量,经统计处理,无显著差异。一仪器与试剂 GFU-202型原子吸收光谱仪;铜空心阴极灯饱和氯化钾溶液 NaOH-硼酸-KCl缓冲溶液:pH 9.0,取0.1MNaOH溶液21.4ml和0.2M硼酸与0.2M KCl混合液25ml混合均匀(0.2M硼酸与0.2M KCl混合液:
Determination of glutamate in MSG content, and more use of optical rotation and neutralization method. Since amino acids can be quantitatively complexed with copper from a copper phosphate precipitate under alkaline conditions, the content of glutamic acid in the indirect determination of copper can be determined by atomic absorption spectrometry.
