目的探讨用核仁组成区嗜银蛋白(AgNORs)染色反映压力超负荷后早期心肌肥大的可能性。方法作心肌AgNORs染色并作图像分析进行相对定量,分析其与心肌肥大进程的关系。结果随着大鼠血压的升高,左室重量指数于手术5d开始显著增大(P<0.05),并持续升高。超微结构显示术后1d心肌细胞肌浆网逐渐扩张,术后5d尤以核周和肌膜下明显。线粒体立体计量显示,仅线粒体数密度N在术后5d时显著增多(P<0.05),与心肌肥大相关;AgNORs平均颗粒数N、颗粒面积和积分光密度IOD在术后1d开始出现显著变化,即N显著升高,A、IOD显著降低,这种改变持续至术后30d。结论AgNORs染色可反映心肌细胞核活性早期变化,是显示心肌肥大启动的敏感形态学方法。
Objective To investigate the possibility that the nucleolar organizer region AgNORs staining early myocardial hypertrophy after pressure overload. Methods Myocardial AgNORs staining and image analysis for relative quantification, and its relationship with the process of cardiac hypertrophy. Results With the increase of blood pressure in rats, the weight index of left ventricle increased significantly (P <0.05) on the 5th day after operation and continued to increase. The ultrastructure showed that the myocardial sarcoplasmic reticulum gradually dilated at 1 day after operation, especially in the peri-nuclear and sub-myometrium at 5 days after operation. Mitochondrial stereological measurements showed that the number density of mitochondria only increased significantly at 5 days after operation (P <0.05), and was correlated with cardiac hypertrophy. The average number of particles of AgNORs N, particle area and integral optical density (IOD) Changes, namely N significantly increased, A, IOD decreased significantly, this change lasted until 30d. Conclusion AgNORs staining can reflect the early changes of myocardial cell nuclear activity and is a sensitive morphological method to show the initiation of cardiac hypertrophy.
