目的:探讨抑癌基因p16的表达与前列腺癌发生、发展的关系,从而从翻译水平研究p16基因与前列腺癌生物学行为的关系.方法:采用免疫组织细胞化学SABC染色法技术、兔抗人pl6多克隆抗体(C-20)和组织抗原修复技术(SanyoEM-715FW微波炉的“慢速烹调、高火力档”15’)对41例前列腺癌标本进行检测p16基因蛋白的表达结果:①在41例前列腺癌组织标本中,蛋白的阳性表达率(为48.8%)较高.提示p16基因的突变或缺失可能在前列腺癌的发生发展中具有重要作用.SABC法免疫组化的结果表明,P16蛋白均表达在前列腺癌腺上皮细胞的细胞质中,大部分呈弥漫均匀分布,部分呈区域性表达.②p16基因蛋白低表达是病理分级高(恶性程度高)、具有侵袭性前列腺癌的重要特征,是其晚期事件.pl6基因蛋白表达率在病理Ⅰ,Ⅱ,Ⅲ级分别为80%,474%,25%,差异显著(P<0.05);在局限于前列腺包膜内的A+B期为65.4%,侵袭包膜外及扩散、转移的C+D期为20%,差异显著(P<O.05);在死亡组为卫2.5%,存活组为61.5%,差异显著(P<O.05),预示检测P16蛋白可作为判断前列腺癌预后的一个指标.结论:p16基因与前列腺癌的发生、发展密切相关;参与前列腺癌的自然病程;p16蛋白的检测是判定前列腺癌预后的一个有用的指
