目的长爪沙鼠真核蛋白延长因子1a2(eukaryotic translation elongation factor 1 alpha 2,Eef1a2)基因的克隆,测序和同源性分析。方法提取长爪沙鼠骨骼肌组织的mRNA,通过同源扩增和cDNA末端快速扩增技术(rapid amplification of cDNA ends,RACE)克隆Eef1a2 cDNA序列全长。通过DANSTAR软件比对长爪沙鼠和人类、大鼠、小鼠Eef1a2的cDNA和氨基酸序列,并分析其同源性。结果长爪沙鼠Eef1a2 cDNA序列全长1812 bp,编码区(coding sequence,CDs)1392 bp,编码463个氨基酸。其核苷酸序列与人类、大鼠和小鼠的同源性分别为88.6%,94.0%和93.8%;长爪沙鼠Eef1a2氨基酸序列与人类、大鼠和小鼠的同源性分别为100%,99.9%和99.9%。结论长爪沙鼠Eef1a2的核苷酸序列和氨基酸序列均与人、大鼠、小鼠高度同源。因此,长爪沙鼠可能是一种人类Eef1a2基因研究的理想模型。
Objective To clone, sequence and homology analysis of eefarya translation elongation factor 1 alpha 2 (Eef1a2) gene. Methods The mRNA of skeletal muscle tissue of Meriones unguiculatus was extracted and the full-length cDNA sequence of Eef1a2 was cloned by homologous amplification and rapid amplification of cDNA ends (RACE). The cDNA and amino acid sequences of Eef1a2 from Meriones unguiculatus and human, rat and mouse were compared by DANSTAR software and their homologies were analyzed. Results The cDNA sequence of Eef1a2 was 1812 bp in length and coding sequence (CDs) was 1392 bp, encoding 463 amino acids. The homology of the nucleotide sequence to human, rat and mouse were 88.6%, 94.0% and 93.8% respectively. The amino acid sequence of Eef1a2 in Meriones unguiculatus was 100 %, 99.9% and 99.9%. Conclusion The nucleotide and amino acid sequences of Eef1a2 are highly homologous to human, rat and mouse. Therefore, Mongolian gerbils may be an ideal model for human Eef1a2 gene research.
