应用不同剂量的氯化镉(0.2、0.4和0.8mg/kgBW)对成年雄性SD大鼠进行腹腔染毒,连续染毒7天后将动物处死,分离睾丸,测定其中金属硫蛋白(MT)含量、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)等抗氧化酶活性和脂质过氧化产物丙二醛(MDA)含量。结果显示,低剂量组MT含量虽有所升高,但与对照组比较差异无显著性,中剂量组MT含量显著升高,高剂量组MT含量反而显著降低。睾丸SOD、GSH-Px等抗氧化酶的活性和MDA含量在低、中剂量组变化不明显,而在高剂量组,SOD和GSH-Px的活性显著降低,MDA含量显著升高。结果揭示,MT对镉所致的睾丸氧化损伤起重要的保护作用,镉的雄性生殖毒性可能与MT及其他抗氧化物质的抗氧化失衡有关。
Adult male SD rats were intraperitoneally treated with different doses of cadmium chloride (0.2, 0.4 and 0.8 mg / kg BW). Animals were sacrificed 7 days after continuous exposure and the testis was isolated. The content of metallothionein (MT) Superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and other antioxidant enzymes activities and lipid peroxidation product malondialdehyde (MDA) content. The results showed that although the MT content of low dose group increased, but no significant difference compared with the control group, MT content was significantly increased in the middle dose group, high dose group MT content was significantly reduced. The activity of SOD and GSH-Px and other antioxidant enzymes in testis and the content of MDA in the low and medium dose groups did not change obviously. However, the activities of SOD and GSH-Px in the high dose group decreased significantly and MDA content increased significantly. The results revealed that MT plays an important protective role in cadmium-induced oxidative damage of testis, and the male reproductive toxicity of cadmium may be related to the antioxidant imbalance of MT and other antioxidants.
