目的初步探讨我国结核分支杆菌的数目可变串联重复位点 (VariableNumberofTandemRepeat ,VNTR)的分布特征及其在基因分型中的应用。方法 选取结核分支杆菌基因组中的 5个VNTR位点 ,采用PCR和琼脂糖凝胶电泳技术 ,并应用BioNumerics(Version3.0 )软件进行处理 ,分析结核分支杆菌DNA中VNTR分布多态性。结果 检测分析的 6 5株结核分支杆菌具有明显的多态性 ,共分成 12个不同的型别 ,其中大部分菌株属于一个型 ,占 6 4 .6 % ,其他菌株呈散在分布。结论 我国结核分支杆菌的VNTR具有明显的多态性。VNTR分型技术具有简便、快速、特异、重复性好等优点 ,可广泛用于结核病的分子流行病学研究。
Objective To investigate the distribution characteristics of Variable Number of Tandem Repeat (VNTR) in Mycobacterium tuberculosis in China and its application in genotyping. Methods Five VNTR loci in Mycobacterium tuberculosis were selected and analyzed by PCR and agarose gel electrophoresis. Bio VNumerics (Version3.0) software was used to analyze VNTR distribution polymorphisms in Mycobacterium tuberculosis. Results Sixty-five Mycobacterium tuberculosis strains were detected and analyzed. There were 12 different genotypes, most of which belonged to one type, accounting for 64.4%. Other strains were scattered. Conclusion VNTR of Mycobacterium tuberculosis in China has obvious polymorphism. VNTR typing technology has the advantages of simple, rapid, specific, good repeatability, which can be widely used in the molecular epidemiology of tuberculosis.
