一氧化氮(NO)是一种胃肠道的抑制性神经递质,可使器官和血管的平滑肌细胞松弛,这一作用的第二信使是环磷酸鸟苷(cGMP)。由于磷酸二酯酶5(PD5)的作用,环磷酸鸟苷被水解而失活。 西地那非是一种用来治疗勃起障碍的磷酸二酯酶抑制剂,通过阻断PD5来阻止NO刺激产生的细胞内cGMP的降解,从而使平滑肌细胞松弛。本研究的目的是调查西地那非对高收缩性食管动力障碍病人和健康人的食管体蠕动性动力和下食管括约肌的影响。
Nitric oxide (NO) is a gastrointestinal inhibitory neurotransmitter that relaxes the smooth muscle cells of organs and blood vessels. The second messenger of this effect is guanosine monophosphate (cGMP). Due to the action of phosphodiesterase 5 (PD5), guanosine monophosphate is inactivated by hydrolysis. Sildenafil, a phosphodiesterase inhibitor used to treat erectile dysfunction, relaxes smooth muscle cells by blocking PD5 to prevent the degradation of intracellular cGMP produced by NO stimulation. The purpose of this study was to investigate the effects of sildenafil on the motility of the esophageal motility and the lower esophageal sphincter in patients with hypersensitive esophageal motility and in healthy individuals.
