目的建立高效液相色谱梯度洗脱法同时测定复方间苯二酚乳膏中间苯二酚和醋酸曲安奈德含量的方法。方法使用ZORBAX Eclipse XDB-C18(4.6 mm×250 mm,5μm),流动相为乙腈(A)-0.5%醋酸铵溶液(B),进行梯度洗脱,0~3 min(25%A),3~5 min(25%A~55%A),5~15 min(55%A),柱温40℃,流速1.0 mL·min-1,检测波长240 nm。结果间苯二酚和醋酸曲安奈德的线性范围分别为:103.18~515.90μg.mL-1(r=1.000 0),1.336~6.680μg.mL-1(r=0.999 4);平均回收率分别为99.3%,99.1%;RSD分别为0.62%,0.73%。结论该方法简便,快速,准确,用于实际样品的测定,结果满意。
OBJECTIVE To establish a method for simultaneous determination of resorcinol and triamcinolone acetonide in compound resorcinol cream by gradient elution with high performance liquid chromatography. Methods The mobile phase consisted of acetonitrile (A) - 0.5% ammonium acetate solution (B) and gradient elution was performed on a ZORBAX Eclipse XDB-C18 (4.6 mm × 250 mm, 5 μm) ~ 5 min (25% A ~ 55% A), 5 ~ 15 min (55% A) at a flow rate of 1.0 mL · min- Results The linear ranges of resorcinol and triamcinolone acetonide were 103.18-515.90μg.mL-1 (r = 1.000 0) and 1.336-6.680μg.mL-1 (r = 0.999 4), respectively. The average recoveries were 99.3% and 99.1%, respectively; RSD was 0.62% and 0.73% respectively. Conclusion The method is simple, rapid and accurate for the determination of actual samples with satisfactory results.
