目的:探讨nm23H1与大肠癌淋巴结转移的关系。方法:利用限制性片段长度多态性、Southernblot和免疫组织化学染色技术(LSAB法),检测80例大肠癌及正常粘膜DNA中nm23H1杂合性缺失(nm23H1LOH)和石蜡组织切片中nm23H1蛋白表达。结果:80例中45例为杂合子,其中LOH14例,9例有淋巴结转移,31例无LOH者,仅6例有淋巴结转移(P<0.01);nm23H1蛋白表达阴性20例中11例有淋巴结的转移,表达阳性25例中4例有淋巴结转移(P<0.01)。nm23H1LOH和nm23+H1蛋白表达阴性者在DukesC/D期较A、B期多见(P<0.05)。LOH和蛋白表达与年龄、性别、肿瘤大小、部位、组织类型无关。结论:nm23H1LOH与大肠癌淋巴结转移呈正相关,蛋白表达与大肠癌淋巴结转移呈负相关;检测大肠癌组织DNA中nm23H1LOH和组织切片中nm23H1蛋白表达,可能成为预测大肠癌淋巴结转移的生物学指标之一。
Objective: To investigate the relationship between nm23H1 and lymph node metastasis of colorectal cancer. METHODS: Using restriction fragment length polymorphism, Southern blot, and immunohistochemical staining (LSAB) to detect nm23H1 heterozygosity loss (nm23H1LOH) and paraffin sections in 80 cases of colorectal cancer and normal mucosal DNA nm23-H1 protein expression. Results: Of the 80 cases, 45 cases were heterozygous, including 14 cases with LOH, 9 cases with lymph node metastasis, 31 cases without LOH, and 6 cases with lymph node metastasis (P<0.01); nm23-H1 protein expression was negative in 20 cases. 11 cases had lymph node metastasis, and 4 of 25 cases showed lymph node metastasis (P<0.01). The negative expression of nm23H1LOH and nm23+H1 was more common in Dukes C/D than in A and B (P<0.05). LOH and protein expression were independent of age, gender, tumor size, location, and tissue type. Conclusion: nm23H1LOH is positively correlated with lymph node metastasis of colorectal cancer. Protein expression is negatively correlated with lymph node metastasis of colorectal cancer. Detection of nm23H1LOH in colorectal tissue DNA and expression of nm23H1 protein in tissue slices may be a predictor of lymph node metastasis in colorectal cancer. One of the biological indicators.
