本文利用交叉免疫电泳技术对诺氏疟原虫可溶性抗原进行了分析。实验系采用100×100×3mm~3琼脂糖玻璃板和含Ca~(2+)的巴比妥缓冲液、第一向电泳所用电压为8V/cm,电泳1小时,第二向电泳所用电压为2V/cm,电泳6—8/小时。电泳后经压片、洗绦、再压片、干燥、染色、脱色和再干燥步骤后观察结果。实验结果表明,作为标准对照的卵白蛋白抗原和人血清抗原分别与相应的抗血清形成许多沉淀峰,而且其抗原的浓度与沉淀峰高度明显相关。在待测抗原的分析中,诺氏疟原虫可溶性表面抗原能与相应的抗血清形成6条沉淀峰,由于所用抗原预先用同位素进行了标记,因而通过放射自显影提高了敏感性和分辨力。
In this paper, soluble immunogen antigen of Plasmodium knowlesi was analyzed by cross-immunoelectrophoresis. The experimental system used 100 × 100 × 3mm ~ 3 agarose glass plates and Ca ~ (2+) -based barbiturate buffer. The first-dimension electrophoresis voltage was 8V / cm, electrophoresis for 1 hour, the second-dimension electrophoresis voltage 2V / cm, electrophoresis 6-8 / hour. After electrophoresis, tabletting, washing, and then tableting, drying, dyeing, decolorization and re-drying steps after the observation. The experimental results show that the standard ovalbumin antigen and human serum antigens, respectively, with the corresponding antiserum many precipitation peaks, and the antigen concentration was significantly correlated with the precipitation peak height. In the analysis of the tested antigens, the soluble surface antigens of Plasmodium normatum can form six precipitation peaks with the corresponding antisera, and the sensitivity and resolving power by autoradiography are improved since the antigens used are previously labeled with isotopes.
