通过蛋白质双向电泳和质谱技术,对低温前后抗寒品种东农冬麦1号和非抗寒品种济麦22的蛋白质组进行了比较分析.结果表明:在pH 4~7范围内,在东农冬麦1号和济麦22分蘖节蛋白表达图谱中发现55个蛋白点在低温胁迫后的表达量与低温前有明显差异,其中47个点在两个品种中均有表达.低温处理后,有23个蛋白点的丰度值变大,东农冬麦1号的绝对丰度高于济麦22;7个蛋白点的丰度值变小,东农冬麦1号的绝对丰度低于济麦22;东农冬麦1号特有8个蛋白点表达量上升.所有上调的蛋白点中东农冬麦1号的表达量是济麦22的2.1~16.5倍,超过4倍以上的点有10个;所有下调的点中东农冬麦1号的表达量是济麦22的0.1~0.4倍.对所有的差异蛋白点进行质谱分析,并通过数据库检索进行蛋白质鉴定,得到了其中51个蛋白的完整肽指纹图谱,这些蛋白中逆境蛋白占15.7%、代谢相关蛋白占27.5%、信号分子占19.6%、未知蛋白占9.8%、其他类蛋白占27.4%.这些蛋白的表达可能对东农冬麦1号抗寒性具有重要作用.
The proteome analysis of Dongnongdongmai 1, a non-cold-tolerant cultivar and Jimai 22, was conducted by means of two-dimensional electrophoresis and mass spectrometry. The results showed that in the range of pH 4-7, The protein expression profiles of 55 protein spots in winter wheat 1 and Jimai 22 tillers were significantly different from those before low temperature, of which 47 spots were expressed in both cultivars.After low temperature treatment, The abundance of 23 protein spots became larger, the absolute abundance of Dongnongdongmai 1 was higher than that of Jimai 22, and the abundance of 7 protein spots became smaller. The absolute abundance of Dongnongdongmai 1 was lower In Jimai 22, the expression of 8 unique protein spots in Dongnongdongmai 1 increased.The expression of Zhongnong Dongmai 1 in all of the up-regulated protein spots was 2.1 ~ 16.5 times of that in Jimai 22, and more than 4 times All of which were down-regulated.The middle Dongnongdongmai 1 was 0.1 ~ 0.4 times of that of Jimai 22. All of the differential protein spots were analyzed by mass spectrometry, and the protein identification was carried out by database search, of which 51 Proteins complete peptide fingerprinting, these proteins adversity protein accounted for 15.7%, metabolic-related protein accounted for 27.5%, signaling molecules 19.6%, accounting for 9.8% of unknown proteins, and other proteins accounted for 27.4%. The expression of these proteins might play an important role in Dongnongdongmai 1 cold resistance.
