目的分析结直肠癌组织中赖氨酰氧化酶(LOX)的表达与患者临床病理特征及预后的关系,并探讨LOX在结直肠癌进展中的作用。方法收集2009年1月至2010年5月在我院确诊并行肿瘤切除的82例结直肠癌患者的癌组织、癌旁组织石蜡标本及临床资料、随访信息。免疫组化染色检测结直肠癌及癌旁组织中LOX、缺氧诱导因子-1α(HIF-1α)、基质金属蛋白酶-2(MMP-2)、MMP-7蛋白的表达情况。分析LOX与患者临床病理参数及预后的关系,进一步探讨LOX与HIF-1α、MMP-2、MMP-7之间的关系。结果结直肠癌组织中LOX表达率较癌旁组织高(P<0.05)。LOX表达阳性的患者肿瘤临床分期更晚、浸润深度更深、淋巴结转移更多(P均<0.05);预后分析显示LOX表达阳性患者的预后较差,COX模型显示LOX表达和TNM分期Ⅲ~Ⅳ期是影响结直肠癌患者预后的独立危险因素(P<0.05)。结直肠癌组织HIF-1α、MMP-2、MMP-7阳性表达率高于癌旁组织(均P>0.05)。相关分析显示LOX表达与HIF-1α、MMP-2蛋白之间存在正相关(P<0.05)。结论 LOX蛋白在结直肠癌组织中表达增强,与结直肠癌的进展有关,且可作为预后预测因子。LOX可能与HIF-1α、MMP-2共同作用促进结直肠癌进展。
Objective To analyze the relationship between the expression of lysyl oxidase (LOX) and clinicopathological features and prognosis in colorectal cancer and to explore the role of LOX in the progression of colorectal cancer. Methods A total of 82 patients with colorectal cancer who had been diagnosed with concurrent tumor resection in our hospital from January 2009 to May 2010 were retrospectively collected. Immunohistochemical staining was used to detect the expression of LOX, HIF-1α, MMP-2 and MMP-7 in colorectal cancer tissues and paracancerous tissues. The relationship between LOX and clinicopathological parameters and prognosis was analyzed, and the relationship between LOX and HIF-1α, MMP-2 and MMP-7 was further explored. Results The expression of LOX in colorectal cancer tissues was higher than that in paracancerous tissues (P <0.05). Patients with positive LOX expression had later clinical stage, deeper invasion and more lymph node metastasis (P <0.05). Prognostic analysis showed that patients with positive LOX expression had a poor prognosis. COX model showed that LOX expression and TNM stage Ⅲ ~ Ⅳ Is an independent risk factor affecting the prognosis of patients with colorectal cancer (P <0.05). The positive rates of HIF-1α, MMP-2 and MMP-7 in colorectal cancer tissues were higher than those in paracancerous tissues (all P> 0.05). Correlation analysis showed that there was a positive correlation between LOX expression and HIF-1α, MMP-2 protein (P <0.05). Conclusion The expression of LOX protein in colorectal cancer tissues is increased, which is related to the progress of colorectal cancer and can be used as prognostic factor. LOX may cooperate with HIF-1α, MMP-2 to promote the progress of colorectal cancer.
