目的:探讨人参皂苷Rg1/ADSCs辅以透明质酸为基质对兔颞下颌关节骨关节病的治疗效果。方法:32只新西兰大白兔随机分为空白组、模型组、对照组、实验组,每组8只。通过关节腔内注射木瓜蛋白酶法建立骨关节病模型,对照组与实验组造模成功2周后开始药物治疗。对照组关节上腔注射人参皂苷Rg1/ADSCs悬液0.6mL,实验组注射透明质酸/人参皂苷Rg1/ADSCs复合物0.6mL,每周1次。各组动物分别于用药第3、9周后处死,每次16只,每组4只。制备标本,行髁突软骨扫描电镜、组织学观察及实时定量PCR检测,采用SPSS17.0软件包对数据进行方差分析及t检验。结果:扫描电镜与组织学观察可见,实验组软骨病变区改善情况优于对照组。与模型组比较,实时定量PCR结果显示,实验组MMP-13表达与空白组相对持平但低于对照组,差异有显著性(P<0.05);实验组TIMP-1表达与空白组相对持平但高于对照组,差异有显著性(P<0.05)。结论:人参皂苷Rg1/ADSCs辅以透明质酸为基质对改善兔颞下颌关节骨关节病有明显作用。
Objective: To investigate the therapeutic effect of ginsenoside Rg1 / ADSCs supplemented with hyaluronic acid on temporomandibular joint osteoarthrosis in rabbits. Methods: 32 New Zealand white rabbits were randomly divided into blank group, model group, control group and experimental group, with 8 in each group. The model of osteoarthropathy was established by intra-articular injection of papain, and the control group and the experimental group were started the drug treatment after 2 weeks of success. In the control group, 0.6 mL of ginsenoside Rg1 / ADSCs suspension was injected into the joint and 0.6 mL of hyaluronic acid / ginsenoside Rg1 / ADSCs complex was injected into the experimental group once a week. The animals in each group were sacrificed after the third and ninth weeks of treatment, each with 16 rats in each group. Specimens were prepared and examined for condylar cartilage by scanning electron microscopy, histological observation and real-time quantitative PCR. SPSS17.0 software package was used for analysis of variance and t-test. Results: Scanning electron microscopy and histological observation showed that the improvement of cartilage lesions in the experimental group was better than that of the control group. Compared with the model group, real-time quantitative PCR results showed that the expression of MMP-13 in the experimental group was relatively flat but lower than that in the control group (P <0.05), while the expression of TIMP-1 in the experimental group was relatively flat Higher than the control group, the difference was significant (P <0.05). Conclusion: Ginsenoside Rg1 / ADSCs supplemented with hyaluronic acid as matrix can significantly improve the temporomandibular joint osteoarthrosis in rabbits.
