目的研究中国甘肃东乡族钙蛋白酶10基因单核苷酸多态性(SNP43)(G/A)位点多态性,探讨其与胰岛素抵抗、2型糖尿病发病的关系。方法应用聚合酶链式反应-限制性片段长度多态性技术对随机选取的东乡族2型糖尿病患者38例和同期体检的85名东乡族对照组钙蛋白酶10基因SNP43(G/A)位点进行多态性分析。采用氧化酶法或放免法测定血糖、胰岛素及血脂水平。结果 2型糖尿病组和对照组基因型及等位基因的频率比较差异无统计学意义;在2型糖尿病组钙蛋白酶10基因SNP43的G/G型与G/A型及A/A型携带者空腹胰岛素、胰岛素抵抗指数相比较,差异均有统计学意义(P<0.05),而空腹血糖、甘油三酯、总胆固醇、高密度脂蛋白胆固醇、低密度脂蛋白胆固醇在2型糖尿病组和对照组3种基因型携带者间相比较,差异均无统计学意义。结论钙蛋白酶10 SNP43(G/A)在中国甘肃东乡族人群中存在基因多态性,该基因多态性与甘肃地区东乡族人群胰岛素抵抗相关。
Objective To investigate the SNP43 (G / A) polymorphism of the calpain 10 gene in Dongxiang, Gansu Province, China, and to explore its relationship with insulin resistance and type 2 diabetes mellitus. Methods The polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method was used to determine the association between the SNP43 (G / A) locus in 38 randomly selected Dongxiang people with T2DM and 85 Dongxiang ethnic controls in the same period. Polymorphism analysis. Oxidation or radioimmunoassay was used to measure blood glucose, insulin and lipid levels. Results There was no significant difference in genotype and allele frequency between type 2 diabetes mellitus group and control group. In type 2 diabetic patients, G / G and G / A type and A / A type carriers of calpain 10 gene SNP43 Fasting insulin and insulin resistance index, the difference was statistically significant (P <0.05), while fasting blood glucose, triglyceride, total cholesterol, high density lipoprotein cholesterol, low density lipoprotein cholesterol in type 2 diabetes group and control There was no significant difference between the three genotype carriers. Conclusion The presence of gene polymorphism of calpain 10 SNP43 (G / A) in Dongxiang ethnic group in Gansu Province of China is related to the insulin resistance in Dongxiang ethnic group in Gansu Province.
