为保持食用菌菌种的优良种性和防止杂菌污染,较长期保藏斜面菌种,我们试用蜡封管口法,效果良好。采用此法无需专门设备、特殊药物,材料易得,操作简便。具体做法如下: 1.培养基的制备。取大型试管(口径为20~25毫米),适当加大培养基中琼脂的比例(占3%),增加注入试管中培养基的数量(占容积的1/4),棉塞要做紧。培养基的配方应根据该菌种生长发育对营养的需求,选择最佳方案,一般多采用马铃薯葡萄糖琼脂培养基(PDA培养基)。 2.接种培养。在已选定的菌株中,挑取茁壮的菌丝为接种体,接种体不要过大(3×3毫米)。在该菌生长最适的温度下进行培养,长满斜面后须继续培养2~3天。 3.剪塞浸蜡。将已培养好的斜面,齐管
