目的研究Survivin基因在儿童白血病中的表达及其意义。方法应用免疫组化PV-9000方法对104例白血病患儿、41例治疗后完全缓解患儿、52例非恶性血液病患儿的骨髓进行检测。结果采用显色强度结合阳性细胞百分比法进行判读。Survivin基因在白血病组中的阳性表达率(56.73%)显著高于完全缓解组(14.63%)和非恶性血液病组(11.54%),但在完全缓解组和非恶性血液病组间没有差异,Survivin基因在急性淋巴细胞白血病组(54.93%)和急性非淋巴细胞白血病组(60.61%)间的阳性表达率亦无差异。结论Survivin基因可能与儿童白血病的发生发展有关,Survivin基因的表达可以作为诊断和观察白血病预后的指标,为白血病的生物治疗提供新思路。
Objective To study the expression of Survivin gene in childhood leukemia and its significance. Methods Immunohistochemistry PV-9000 was used to detect the bone marrow of 104 children with leukemia, 41 children with complete remission after treatment and 52 children with non-hematologic malignancies. Results using color intensity combined with the percentage of positive cells for interpretation. The positive expression rate of Survivin gene in leukemia group was significantly higher than that in complete remission group (14.63%) and non-hematologic malignancies group (56.73%), but there was no difference between complete remission group and non-hematologic malignancies group There was no difference in the positive rates of Survivin between acute lymphoblastic leukemia (54.93%) and acute non-lymphocytic leukemia (60.61%). Conclusions Survivin gene may be involved in the development of childhood leukemia. The expression of Survivin gene may serve as an index for diagnosis and observation of the prognosis of leukemia and provide new ideas for the biotherapy of leukemia.
