目的:建立芦荟大黄素在大鼠血浆中的固相萃取-高效液相色谱(SPE-HPLC)分析方法,研究大鼠体内的药代动力学过程。方法:SD大鼠,按50和20 mg.kg-1剂量灌胃给药,不同时间点尾静脉取血,以SPE处理血浆样品,用HPLC内标法测定血药浓度,甲醇-0.1%磷酸水(70∶30)为流动相,柱温为30℃,检测波长254 nm,以DAS2.0软件计算药代动力学参数。结果:血浆中芦荟大黄素在0.016~19.53μg.mL-1(r=0.998 1)范围内线性关系良好,提取回收率为88.7%~94.6%,最低检测限为0.008μg.mL-1,定量限为0.016μg.mL-1,日内和日间精密度RSD均<6%。结论:芦荟大黄素在大鼠体内呈二室模型,分布广泛。建立的测定大鼠体内芦荟大黄素含量的方法具有良好的准确度、精密度以及专属性,适用于芦荟大黄素在大鼠体内的药代动力学研究。
OBJECTIVE: To establish a SPE-HPLC method for the determination of aloe-emodin in rat plasma and to study the pharmacokinetics of aloe-emodin in rats. Methods: Sprague - Dawley rats were drenched with 50 and 20 mg · kg -1 of gavage. Blood samples were taken from the tail vein at different time points. The plasma samples were processed by SPE. The plasma concentrations of methanol - 0.1% phosphoric acid Water (70:30) as the mobile phase, the column temperature was 30 ℃, the detection wavelength was 254 nm, and the pharmacokinetic parameters were calculated by DAS2.0 software. Results: Aloe-emodin in plasma showed good linearity in the range of 0.016 ~ 19.53μg.mL-1 (r = 0.998 1) with the recovery of 88.7% ~ 94.6% and the lowest detection limit of 0.008μg.mL-1 The limit was 0.016μg.mL-1, intra-day and inter-day precision RSD <6%. Conclusion: Aloe-emodin has a two-compartment model in rats and is widely distributed. The established method of determination of aloe-emodin in rat has good accuracy, precision and specificity, and is suitable for the study of pharmacokinetics of aloe-emodin in rats.
