目的:探讨骨髓间充质干细胞(BMSCs)联合柴胡疏肝散对心肌梗死大鼠心肌组织的保护作用。方法:采用全骨髓贴壁法培养BMSCs。经结扎冠状动脉制备大鼠心肌梗死(AMI)模型。SD大鼠随机分为假手术组、AMI组(模型组)、BMSCs组、CHSG-L组(柴胡疏肝散小剂量+BMSCs移植)、CHSG-M组(柴胡疏肝散中剂量+BMSCs移植)、CHSG-H组(柴胡疏肝散大剂量+BMSCs移植)。BMSCs细胞悬液直接注入梗死区边缘心肌组织;中药或生理盐水经灌胃给予。各组干预7 d后,生化法测定大鼠血清超氧化物歧化酶(SOD)与丙二醛(MDA)含量;ELISA法检测大鼠血清低氧诱导因子(HIF-1α)表达;免疫组化法观察心肌血管内皮生长因子(VEGF)蛋白表达。结果:与模型组和BMSCs组比较,各联合治疗组SOD升高、MDA降低,血清HIF-1α水平和心肌细胞VEGF蛋白表达升高;且柴胡疏肝散剂量越大,作用越显著。结论:BMSCs移植联合柴胡疏肝散可一定程度上保护心肌梗死大鼠心肌组织。
Objective: To investigate the protective effect of bone marrow mesenchymal stem cells (BMSCs) combined with Chaihu Shugan San on myocardial tissue in rats with myocardial infarction. Methods: BMSCs were cultured with whole bone marrow adherent method. Rat myocardial infarction (AMI) model was established by ligation of coronary artery. SD rats were randomly divided into sham operation group, AMI group (model group), BMSCs group, CHSG-L group (small dose of Chaihu Shugan San + BMSCs transplantation), CHSG- BMSCs transplantation), CHSG-H group (high dose of Chaihu Shugan Powder + BMSCs transplantation). BMSCs cell suspension injected directly into the infarct border myocardial tissue; Chinese medicine or saline given by gavage. Serum levels of superoxide dismutase (SOD) and malondialdehyde (MDA) were measured by biochemical methods after 7 d intervention in each group. The expression of HIF-1α was detected by ELISA. Method to observe the expression of vascular endothelial growth factor (VEGF) protein. Results: Compared with the model group and the BMSCs group, the SOD and MDA of the combined treatment group were increased, the level of serum HIF-1α and the expression of VEGF protein in myocardial cells were increased, and the greater the dose of Chaihu Shugan Powder was. Conclusion: BMSCs transplantation combined with Chaihu Shugan San can protect myocardium from myocardial infarction to some extent.
