目的:建立多波长高效液相色谱法,同时测定四神丸中五味子醇甲、五味子甲素、五味子乙素、补骨脂素、异补骨脂素、去氢二异丁香酚、吴茱萸次碱7个有效成分的含量。方法:采用Diamond C18色谱柱(4.6 mm×250 mm,5μm),柱温30℃,以乙腈-0.1%磷酸水溶液为流动相梯度洗脱(0 min,30%A;10 min,40%A;30 min,50%A;60 min,90%A;61~70 min,30%A),检测波长为220 nm(五味子醇甲、五味子甲素、五味子乙素)、280 nm(补骨脂素、异补骨脂素、去氢二异丁香酚)和345 nm(吴茱萸次碱)。结果:7个有效成分达到完全分离;线性关系良好;加样回收率在94.8%~104.1%。结论:建立的方法准确可靠,可用于四神丸的质量控制。
OBJECTIVE: To establish a multi-wavelength high performance liquid chromatography (HPLC) method for the simultaneous determination of Schizandrin A, Schizandrin B, Schizandrin B, psoralen, isopsoralen, dehydrogenated diisophenol and rutaecarpine 7 active ingredients content. Methods: The mobile phase was eluted with a gradient of acetonitrile - 0.1% phosphoric acid (0 min, 30% A; 10 min, 40% A) using a Diamond C18 column (4.6 mm × 250 mm, The detection wavelength was 220 nm (Schizandrin A, Schisandrin A, Schisandrin B), 280 nm (Psoralen, 30 min, 50% A; 60 min, 90% , Isopsoralen, dehydro-iso-eugenol) and 345 nm (rutaecarpine). Results: The seven active ingredients were completely separated. The linearity was good. The average recoveries were 94.8% -104.1%. Conclusion: The established method is accurate and reliable and can be used for quality control of Sishen Pills.
