观察盐酸川芎嗪(TMPH)30mg·kg-1iv后大白鼠心、肝、脾、肾、脑、肌肉、睾丸和血浆等组织中药物浓度的经时变化。统计矩分析结果表明:肝脏摄取率最高,AUC为247.9mg·h-1·L-1,其它依次为心脏、脾、脑、睾丸、肺、肾、肌肉、血浆;各组织药物消除的速度依次为:脾、血浆、肌肉、肺、脑、肾、肝、睾丸及心脏,心脏消除最慢,MRT为4.27h。我们认为:药物体内的脏器分布用统计矩法算出的AUC、MRT,再结合Cmax、Tmax等参数进行描述较为合理。
The changes of the drug concentration in the tissues of heart, liver, spleen, kidney, brain, muscle, testis and plasma of rats after 30 mg · kg-1 iv tetramethylpyrazine hydrochloride (TMPH) were observed. The results of statistical moment analysis showed that the highest liver uptake rate was AUC 247.9mg · h-1 · L-1, followed by heart, spleen, brain, testis, lung, kidney, muscle and plasma; Followed by: spleen, plasma, muscle, lung, brain, kidney, liver, testis and heart, the slowest cardiac elimination, MRT was 4.27h. In our opinion, it is more reasonable to describe the distribution of internal organs in the body using the AUC, MRT calculated by the statistical moment method and the parameters such as Cmax and Tmax.
