目的:研究黄芪葛根汤对实验性糖尿病心肌病变的干预作用及机制。方法:采用高脂饲料喂养和链脲佐菌素腹腔注射的方法建立2型糖尿病大鼠心肌病变模型,观察黄芪葛根汤对血糖、胰岛素、心肌纤维化、转化生长因子-β1(TGF-β1)及其信号蛋白Smad3表达的影响;mRNA表达采用RT-PCR方法检测,蛋白表达采用免疫组化方法。结果:黄芪葛根汤可显著降低糖尿病大鼠空腹血糖、胰岛素,改善胰岛素抵抗,减少心肌羟脯氨酸含量抑制胶原增生,降低心脏指数,并显著抑制心肌TGF-β1/Smad3 mRNA和蛋白的强表达。结论:黄芪葛根汤可以通过调控TGF-β1/Smad3通路显著干预实验性糖尿病心肌纤维化病变。
Objective: To investigate the intervention effect and mechanism of Astragalus Pueraria Decoction on experimental diabetic cardiomyopathy. Methods: Cardiomyopathy models of type 2 diabetic rats were established by intraperitoneal injection of high fat diet and streptozotocin (STZ). The effects of Radix Puerariae decoction on blood glucose, insulin, myocardial fibrosis, transforming growth factor-β1 (TGF-β1) And its signal protein Smad3 expression; mRNA expression was detected by RT-PCR, protein expression by immunohistochemistry. Results: Astragalus Pueraria Decoction could significantly decrease fasting blood glucose and insulin, improve insulin resistance, decrease hydroxyproline content in myocardium, inhibit collagen hyperplasia, decrease cardiac index, and significantly inhibit the expression of TGF-β1 / Smad3 mRNA and protein in myocardium . Conclusion: Astragalus Pueraria Decoction can significantly interfere the pathological changes of experimental diabetic fibrosis through regulating TGF-β1 / Smad3 pathway.
