目的p16/CDKN2基因是新近发现的肿瘤抑制基因,已有研究表明该基因在许多肿瘤出现缺失、突变或重排.胃癌细胞系中有高频率纯合子缺失,应用Southernblot和SSCP技术在胃癌手术标本中未发现有该基因的缺失.本文首次研究胃癌中p16/CDKN2基因在蛋白水平的表达.方法应用蛋白A金探针免疫组化技术对30例胃癌和20例正常胃粘膜标本的p16进行分析.结果所有正常对照组均有p16表达,其阳性细胞百分率为7255%±1722%;而胃癌中的阳性细胞百分率为5490%±2890%,明显低于正常对照(P<005).在肿瘤病例中有2333%(7/30)p16表达减低,10%(3/30)未表达p16.此外有1例呈过度表达.结论胃癌中有p16的异常表达,免疫组化技术可能较准确地反映p16的表达,更适合于临床研究.
Objective The p16/CDKN2 gene is a recently discovered tumor suppressor gene. Previous studies have shown that this gene is missing, mutated, or rearranged in many tumors. There was a high frequency of homozygous deletion in gastric cancer cell lines. No Southern blot and SSCP techniques were used to detect the loss of this gene in surgical specimens of gastric cancer. This paper first studied the expression of p16/CDKN2 gene in gastric cancer. Methods Protein A gold probe immunohistochemistry was used to analyze p16 in 30 cases of gastric cancer and 20 cases of normal gastric mucosa. Results The p16 expression was observed in all normal control groups. The percentage of positive cells was 7255%±1722%. The percentage of positive cells in gastric cancer was 5490%±2890%, which was significantly lower than that of normal controls. <005). In tumor cases, 2333% (7/30) p16 expression decreased, and 10% (3/30) did not express p16. In addition, 1 case was over-expressed. Conclusion There is abnormal expression of p16 in gastric cancer. Immunohistochemistry may accurately reflect the expression of p16, which is more suitable for clinical research.
