对结球甘兰(Brassica olerasea L.var.capitara)自交不亲和系材料的下胚轴、子叶及花茎切段等进行离体培养快速繁殖和再生植株的遗传稳定性等的探讨。经过连续4代培养后,“黄苗”自交不亲和系材料在 MS+BA(4mg/l)的培基养上获得了12267/90天的繁殖率;经过12代的连续培养后,“上黑”自交不亲和系材料在 MS+BA(2mg/l)的培养基上,获得了84983926/365天的紧殖率,其繁殖系数达常规法繁殖系数的18倍。两种材料均在不含任何激素的 MS 培养基上获得了诱导生根最佳效果。幼苗移栽成活率达100%。离体培养的再生植株具有良好的遗传稳定性。
Hypocotyls, cotyledons and rhizome sections of Brassica oleracea L.var.capitara self-incompatible materials were dissected in vitro and the genetic stability of regenerated plants were discussed. After 4 generations of continuous culture, the reproductive rate of 12267/90 days was obtained on culture medium of MS + BA (4mg / l) by “Huangmiao” selfing incompatible material. After 12 generations of continuous culture, The “black” self-incompatible material obtained a yield of 84983926/365 days on MS + BA (2 mg / l) medium with a multiplication factor of 18 times that of the conventional method. Both materials gave the best induction of rooting on MS medium without any hormones. Seedling transplanting survival rate of 100%. Regenerated plants in vitro have good genetic stability.
