以茶树品种‘迎霜’为实验材料,从中分离获得2个编码HSF转录因子的基因。进化分析显示,这2个转录因子分别属于HSF转录因子家族的A5和A4亚族,分别命名为CsHsfA5和CsHsfA4。序列分析显示,CsHsfA5和CsHsfA4的开放阅读框分别为1 440和1 224 bp,分别编码479和407个氨基酸,均含有HSF转录因子保守结合域。CsHsfA5和CsHsfA4均属于亲水性蛋白,具有3个α-螺旋和4个β-折叠的三级结构。实时定量PCR表明,CsHsfA5和CsHsfA4基因在不同温度胁迫下均能在3个不同茶树品种‘迎霜’、‘安吉白茶’和‘云南十里香’中诱导表达,且表达量呈现差异。
The tea tree variety Ying Ying was used as experimental material, and two genes encoding HSF transcription factor were isolated from them. Phylogenetic analysis showed that these two transcription factors belong to the A5 and A4 subfamilies of the HSF transcription factor family, respectively named CsHsfA5 and CsHsfA4. Sequence analysis showed that the open reading frames of CsHsfA5 and CsHsfA4 were 1 440 and 1 224 bp, encoding 479 and 407 amino acids, respectively, and all contained the conserved binding domain of HSF transcription factor. Both CsHsfA5 and CsHsfA4 are hydrophilic proteins with three α-helices and four β-sheet tertiary structures. Real-time quantitative PCR showed that CsHsfA5 and CsHsfA4 genes were induced in three different tea varieties ’Yingshuang’, ’Anji Baicha’ and ’Yunnan Shilouxiang’ under different temperature stress, and the expression levels were different.
