目的:通过观察益气活血复方对心衰大鼠心肌组织mi-CK mRNA及蛋白表达的影响,从能量代谢角度研究中药在慢性心衰治疗中的作用。方法:本实验以冠脉结扎法配合力竭式游泳、减食等方法造成大鼠慢性心衰动物模型,对造模成功大鼠分为模型组、西药组、中药组,没有进行左冠脉结扎手术的假手术大鼠为正常组。检测各组大鼠±dp/dt max、心肌组织mi-CK mRNA及蛋白表达水平。结果:与模型组比较,各药物组±dp/dt max、mi-CK mRNA及蛋白表达均升高,差异有统计学意义(P<0.05)。与西药组比较,中药组±dp/dt max升高明显,差异有统计学意义(P<0.05)。在mi-CK mRNA及蛋白表达方面,两治疗组间比较,差异无统计学意义(P>0.05)。结论:益气活血复方可以增加心衰大鼠心肌mi-CK mRNA及蛋白的表达、改善ATP转运穿梭效率,起到修复心衰心肌能量代谢的作用。
OBJECTIVE: To observe the effects of Yiqi Huoxue Compound on mi-CK mRNA and protein expression in myocardial tissue of heart failure rats and to study the role of traditional Chinese medicine in the treatment of chronic heart failure from the perspective of energy metabolism. Methods: The model of chronic heart failure was induced by coronary ligation combined with exhaustive swimming, diet reduction and other methods. The successful model rats were divided into model group, western medicine group and traditional Chinese medicine group without left coronary artery Ligation surgery of sham rats for the normal group. The ± dp / dt max and the expression of mi-CK mRNA and protein in myocardium were detected. Results: Compared with the model group, the ± dp / dt max and the expression of mi-CK mRNA and protein in each drug group were significantly increased (P <0.05). Compared with western medicine group, the ± dp / dt max of traditional Chinese medicine group increased obviously, the difference was statistically significant (P <0.05). In mi-CK mRNA and protein expression, there was no significant difference between the two treatment groups (P> 0.05). Conclusion: Yiqi Huoxue Compound can increase the expression of mi-CK mRNA and protein in cardiac muscle of CHF rats, improve the efficiency of ATP transport and shuttle, and play an important role in repairing myocardial energy metabolism in CHF rats.
