目的探讨透明质酸酶(Hyaluronidase,Hyase)抑制剂4′,5,7-三羟基黄酮,芹菜配基(Apigenin)对人乳腺癌细胞生长、增殖及侵袭潜力的影响。方法通过体外侵袭模型观察Apigenin对人乳腺癌细胞穿透Matrigel(人工基底膜)能力的影响;MTT法和流式细胞仪检测Apigenin对人乳腺癌细胞株生长和增殖的影响。结果8株人乳腺癌细胞的侵袭潜力存在明显差异,加用Apigenin后其侵袭潜力均明显降低(P<0.05),并与Apigenin存在量效关系;Apigenin对MDA-MB-435S和T47D细胞株的生长抑制作用均呈浓度依赖性;流式细胞仪显示Apigenin阻滞人乳腺癌细胞株MDA-MB-231和ZR-75-30生长于G2/M期(P<0.05)。结论Apigenin可抑制人乳腺癌细胞的增殖和Hyase的表达,并可降低人乳腺癌细胞株的侵袭潜能。
Objective To investigate the effects of hyaluronidase (Hyase) inhibitor 4 ’, 5,7-trihydroxyflavone and Apigenin on the growth, proliferation and invasion potential of human breast cancer cells. Methods The effect of Apigenin on the ability of human breast cancer cells to penetrate Matrigel (artificial basement membrane) was observed by in vitro invasion assay. The effect of Apigenin on the growth and proliferation of human breast cancer cell lines was detected by MTT assay and flow cytometry. Results The invasive potential of 8 human breast cancer cells was significantly different. The addition of Apigenin significantly decreased the invasive potential (P <0.05) and had a dose-dependent relationship with Apigenin. The effect of Apigenin on MDA-MB-435S and T47D cell lines Growth inhibition was concentration-dependent. Flow cytometry showed that Apigenin inhibited the growth of human breast cancer cell lines MDA-MB-231 and ZR-75-30 at G2 / M phase (P <0.05). Conclusion Apigenin can inhibit the proliferation of human breast cancer cells and Hyase expression, and reduce the invasive potential of human breast cancer cell lines.
