[目的]研究稀土氧化钕粉尘对大鼠肺灌洗液(BALF)中细胞总数和蛋白含量的动态变化。[方法]选用SPF级健康成年雄性SD大鼠195只,体重(200±16)g,采用气管非暴露插管灌注法染尘。不同染毒时间组分别采用100 mg/kg稀土氧化钕粉尘悬液0.8 mL和0.9%生理盐水0.8 mL一次性染尘,分别于第3、7、14、21、28天各处死15只实验组大鼠和10只对照组大鼠;不同剂量组按25、50、100、150 mg/kg剂量0.8 mL一次性染尘,在染毒第28天处死各实验组大鼠15只,对照组处死10只(由于实验组染毒时间均为28 d,所以设同一对照组为不同染毒剂量组中实验组对照)。取大鼠支气管肺泡灌洗液,用白细胞计数板在显微镜下对细胞总数进行计数。ELISA法分别测定不同染毒时间组和不同染毒剂量组大鼠BALF中上清液和巨噬细胞蛋白的含量。[结果]不同染毒时间组和不同染毒剂量组中实验组大鼠较对照组大鼠BALF中细胞总数均升高(P<0.05);且在各染毒时间组中细胞总数出现先增加后减少,在第14天达到最低值后又呈增加的变化趋势,但均高于对照组(P<0.05)。在不同染毒剂量组中细胞总数随着染毒剂量增加而逐渐增多,细胞总数均高于对照组(P<0.05)。不同染毒时间组中实验组大鼠较对照组大鼠BALF上清和巨噬细胞中的蛋白含量均明显增高(P<0.01)。不同染毒剂量组中实验组大鼠较对照组大鼠BALF上清液和巨噬细胞中的蛋白含量均明显增高(P<0.01)。不同染毒时间组大鼠BALF上清液和巨噬细胞蛋白含量出现先增加后减少,在第14天达到最低值后又呈增加的变化趋势,但均高于对照组(P<0.05)。不同染毒剂量组中大鼠BALF上清液和巨噬细胞蛋白含量随着剂量增加而逐渐增多,蛋白含量均高于对照组(P<0.05)。[结论]稀土氧化钕粉尘进入大鼠肺组织后,BALF中细胞总数以及上清液和巨噬细胞中蛋白含量均发生了明显的变化。
[Objective] To investigate the dynamic changes of total cellular and protein contents in rat lung lavage fluid (BALF) with rare earth neodymium oxide dust. [Methods] A total of 195 SPF healthy adult male Sprague-Dawley (SD) rats weighing 200 ± 16 g were selected to be exposed to dust by intratracheal intubation. The different exposure time groups were treated with 100 mg / kg rare earth neodymium oxide dust suspension 0.8 mL and 0.9% saline 0.8 mL disposable dust, 15 experimental groups were sacrificed on the 3rd, 7th, 14th, 21st and 28th day respectively Rats and 10 control rats; different doses of 25,50,100,150 mg / kg dose of 0.8 mL of a single dose of dust on the 28th day of the rats were killed in the experimental group of 15 rats were sacrificed in the control group 10 (due to experimental group exposure time is 28 d, so the same control group for different exposure dose group experimental group control). Rat bronchoalveolar lavage fluid was taken and the total number of cells was counted under a microscope using a white blood cell counting plate. The contents of supernatant and macrophage protein in BALF of rats in different exposure time and exposure dose groups were determined by ELISA. [Results] The total number of cells in BALF of rats in experimental group and control group were significantly increased (P <0.05), and the total number of cells in each group increased first (P <0.05). After reaching the lowest value on the 14th day, it showed an increasing trend, but both were higher than the control group (P <0.05). The total number of cells in different dose groups increased gradually with the dose increasing, the total number of cells were higher than the control group (P <0.05). The content of protein in BALF supernatant and macrophages of experimental rats in different exposure time groups were significantly higher than that in control rats (P <0.01). The contents of protein in BALF supernatant and macrophages in experimental group were significantly higher than those in control group in different dose groups (P <0.01). The contents of protein in BALF supernatant and macrophages increased first and then decreased in different exposure time groups, and then increased at the 14th day, but both were higher than those in the control group (P <0.05). The contents of protein in BALF supernatant and macrophage of rats in different dose groups increased gradually with the increase of dose, and the contents of protein were higher than those of control group (P <0.05). [Conclusion] The total number of cells in BALF and the content of protein in supernatant and macrophages changed obviously after neodymium oxide dust entering the lung tissue of rats.
