目的探讨提纯人胆肝细胞再生因子(HHGF)方法及对小鼠中毒性肝炎的治疗作用。方法人胚肝细胞裂解物加热后分别经稳膜;超滤,再经DEAE离子交换层析和FRLCmonoQ阴离子交换色谱,最后经SDS-PAGE分析.用3H-TdR渗入法在人胚肝细胞和SMMC-7721人肝癌细胞株中检测HHGF活性.尾静脉注射HHGF治疗小鼠CCl4,中毒性肝炎。结果纯化的HHGF较纯化前相对活性提高2万倍.HHGF可使小鼠CCl4中毒性肝炎升高的ALT和SBIL在96h时接近正常,死亡率从对照组60%降至为0。并有促进肝细胞DNA合成作用.组织学检查HHGF有促进肝细胞再生和治疗肝损伤作用.结论HHGF对促进肝细胞再生、减轻肝功能损伤有明显作用
Objective To investigate the method of purifying human hepatocyte regeneration factor (HHGF) and its therapeutic effect on toxic hepatitis in mice. Methods Human embryonic hepatocyte lysate was heated to stabilize membrane, respectively. After ultrafiltration, DEAE ion exchange chromatography and FRLCmonoQ anion exchange chromatography were carried out, and finally analyzed by SDS-PAGE. HHGF activity was detected by 3H-TdR permeation in human embryonic hepatocytes and SMMC-7721 human hepatoma cell lines. Treatment of mouse CCl4, toxic hepatitis by tail vein injection of HHGF. Results The purified HHGF had a relative 20,000-fold increase in relative activity before purification. HHGF raised the level of ALT and SBIL in mice with CCl4 toxic hepatitis to nearly normal at 96h, and the mortality rate dropped from 60% to 0 in the control group. And to promote DNA synthesis of liver cells. Histological examination HHGF can promote liver cell regeneration and treatment of liver injury. Conclusion HHGF has a significant effect on promoting liver cell regeneration and reducing liver damage
