目的:建立大蒜药材高效薄层色谱指纹图谱,并对不同地理居群大蒜药材的指纹图谱进行比较分析。方法:采用高效薄层硅胶G板,以正丁醇-正丙醇-冰醋酸-水(3∶1∶1∶1)为展开剂,以0.2%茚三酮溶液显色后,在日光灯下检视并照相,并在500 nm处对各轨道扫描吸收光谱后获得薄层色谱指纹图谱,经指纹图谱系统解决方案软件生成共有模式,进行相似度分析。结果:筛选和优化了高效薄层色谱条件,斑点清晰,分离度好;相似度分析表明,不同地理居群的大蒜药材在药用活性成分相对含量上有不同程度的差异,大致可分为5类。结论:建立的高效薄层色谱指纹图谱,为大蒜药材指纹图谱的研究奠定了方法学基础,可以作为不同来源大蒜药材质量控制的重要依据。
OBJECTIVE: To establish a HPLC fingerprint of garlic medicinal herbs and to compare and analyze the fingerprints of garlic medicinal materials of different geographical populations. Methods: High-performance thin-layer silica gel G plate was used with n-butanol-n-propanol-glacial acetic acid-water (3:1:1:1) as developing solvent and developed with 0.2% ninhydrin solution. After viewing and photographing, and scanning the absorption spectra of the orbits at 500 nm, TLC fingerprints were obtained and the common mode was generated by the fingerprint system solution software for similarity analysis. Results: The conditions of high performance thin layer chromatography were identified and optimized, the spots were clear and the resolution was good. Similarity analysis showed that garlic medicinal materials of different geographical populations had different degrees of relative content of medicinal active ingredients, which could be divided into 5 class. Conclusion: The established TLC fingerprinting has laid a foundation for the study of fingerprinting of medicinal herbs in garlic and can be used as an important basis for the quality control of garlic medicinal materials from different sources.
