目的 :研究催乳素 (PRL)对体外单独培养的Graves病 (GD)外周血单个核细胞的活化及增殖有无直接影响。方法 :提取 12例GD患者和 8例正常健康对照的外周血单个核细胞 ,单用含不同浓度羊催乳素或植物血凝素的培养液进行体外培养。并利用流式细胞技术等测定其表面CD2 5的表达 (反映活化状态 )或细胞内DNA含量以计算增殖指数 (反映增殖状态 )。结果 :GD患者和正常人的外周血单个核细胞经 12 .5 10 0 0ng/ml羊催乳素作用后 ,其CD4+ CD2 5 + 细胞百分率、CD8+ CD2 5 + 细胞百分率及增殖指数均无明显变化 (P均 >0 .0 5 )。所设阳性对照植物血凝素 (5 μg/ml)能直接刺激这两种来源的单个核细胞发生活化、增殖 ,使上述各项指标均显著增高 (P均<0 .0 1)。结论 :催乳素不能刺激体外单独培养的GD外周血单个核细胞发生活化及增殖
Objective: To investigate whether prolactin (PRL) has a direct effect on the activation and proliferation of peripheral blood mononuclear cells in Graves’ disease (GD) cultured in vitro. Methods: Peripheral blood mononuclear cells from 12 patients with GD and 8 healthy controls were collected and cultured in vitro with culture medium containing different concentrations of prolactin or phytohemagglutinin. And using flow cytometry to determine the surface CD2 5 expression (reflecting the activation state) or intracellular DNA content to calculate the proliferation index (reflecting the proliferation status). Results: There was no significant change in CD4 + CD25 + cell percentage, CD8 + CD25 + cell percentage and proliferation index of peripheral blood mononuclear cells of GD patients and normal subjects after they were treated with 12.5 × 10 ^ 0 ng / ml sheep prolactin P> 0.05). The positive control lectin (5 μg / ml) can directly stimulate the activation and proliferation of mononuclear cells in these two kinds of sources, so that the above indexes were significantly increased (all P <0.01). CONCLUSION: Prolactin can not stimulate the activation and proliferation of peripheral blood mononuclear cells in vitro cultured in vitro
