目的探讨刺五加注射液体外诱导大鼠骨髓基质细胞分化为神经样细胞的最佳浓度。方法体外密度梯度法分离培养骨髓基质细胞,以不同浓度刺五加注射液对其进行体外诱导,用MTT法检测各组细胞生长情况,用免疫细胞化学方法计算各组神经样细胞阳性数加以比较。结果用不同浓度刺五加注射液诱导后均可表现为典型的神经样细胞形态,其中0.3mg/ml组和0.4mg/ml组细胞生长情况与免疫组化结果均与其余各组差异有统计学意义(P<0.05或P<0.01)。结论刺五加注射液体外诱导成年大鼠骨髓基质细胞分化为神经样细胞以0.3~0.4mg/ml浓度最佳。
Objective To investigate the optimum concentration of rat bone marrow stromal cells differentiated into neuron-like cells induced by Acanthopanax injection. METHODS: Bone marrow stromal cells were isolated and cultured in vitro with density gradient method. In vitro induction was performed with different concentrations of Acanthopanax senticosus injection. The growth of cells in each group was detected by MTT assay. The number of neuron-like cells in each group was calculated by immunocytochemistry and compared. . Results After induction with different concentrations of Acanthopanax Injection, the neuron-like cell morphology was typical. The cell growth and immunohistochemistry results in the 0.3 mg/ml group and the 0.4 mg/ml group were statistically different from the rest of the groups. Significance (P<0.05 or P<0.01). Conclusion Acupuncture plus injection of liquid induces the differentiation of adult rat bone marrow stromal cells into neuron-like cells with a concentration of 0.3-0.4 mg/ml.
