目的观察不同浓度重楼皂苷对人结肠癌细胞凋亡的影响,并研究诱导结肠癌细胞凋亡的分子作用机制:明确重楼皂苷是否对IL-6/JAK-STAT3凋亡调控分子通路产生影响;寻找其诱导结肠癌细胞凋亡的相关信号转导通路或靶点。方法采用不同浓度的重楼皂苷(10,20,40,80μg/ml)分别作用于体外培养的结直肠癌SW480细胞株,在荧光显微镜下观察重楼总皂苷对SW480细胞形态的影响;ELISA法检测血清IL-6的水平;免疫印迹法检测JAK-STAT3蛋白表达水平。结果经重楼皂苷处理后,Hoechst33258染色可见典型的凋亡形态学改变;血清中IL-6分泌明显下降(P<0.05);其信号转导和转录活化因子STAT3降低。结论重楼皂苷能显著诱导结直肠癌SW480细胞产生凋亡,其机制可能为通过下调IL-6的分泌而抑制IL-6/STAT3信号通路的表达。
OBJECTIVE: To observe the effect of different concentrations of Rhizoma Paridis on the apoptosis of human colon cancer cells and to study the molecular mechanism of apoptosis induced by colon cancer cells: To investigate whether the effect of polysalins on the apoptosis pathway of IL-6 / JAK-STAT3 To find out the related signal transduction pathway or target that induce the apoptosis of colon cancer cells. Methods Different concentrations of resplenomide (10, 20, 40, 80μg / ml) were used to in vitro cultured colorectal cancer SW480 cell line, the fluorescence microscope was used to observe the effect of Rhizoma Paridis total saponin on the morphology of SW480 cells. The level of IL-6 in serum was detected. The expression of JAK-STAT3 protein was detected by Western blotting. Results After HAPT treatment, Hoechst33258 staining showed a typical morphological changes of apoptosis. The secretion of IL-6 in serum was significantly decreased (P <0.05), and the signal transducers and activators of transcription STAT3 were decreased. Conclusions Saponin can significantly induce the apoptosis of colorectal cancer SW480 cells, and its mechanism may be through inhibiting the secretion of IL-6 and inhibiting the expression of IL-6 / STAT3 signaling pathway.
