目的:比较大脑基底节区和皮质区对大脑局部缺血/再灌流损伤反应的敏感性。方法:运用免疫组化和酶组化方法对36只SD大鼠局部脑缺血区血管内皮细胞粘附分子VCAM-1的表达和单核/巨噬细胞的浸润概况进行了比较研究。结果:大脑皮质缺血区血管内皮细胞VCAM-1的表达发生于脑缺血1h,表达高峰发生于脑缺血1h/再灌流16h;而在缺血侧基底节区,其表达高峰则发生在脑缺血1h/再灌流4h。单核/巨噬细胞的浸润,在皮质缺血区发生在脑缺血1h/再灌流8h,再灌流16h达到高峰;在缺血侧基底节区,其浸润发生在脑缺血1h,浸润高峰则在脑缺血1h/再灌流8h。结论:大脑基底节区血管内皮细胞粘附分子的表达和单核/巨噬细胞的浸润均较皮质区显著。提示基底节区对脑缺血的反应比皮质区敏感。
OBJECTIVE: To compare the sensitivity of cerebral basal ganglia and cortex to ischemic / reperfusion injury in the brain. Methods: The expressions of VCAM-1 and infiltration of monocyte / macrophage were detected by immunohistochemistry and enzyme-linked immunosorbent assay (ELISA) in 36 ischemic brain regions of rats. Results: The expression of VCAM-1 in the cerebral cortex ischemic area vascular endothelial cells occurred in 1h of cerebral ischemia, and peaked at 1h after cerebral ischemia / reperfusion 16h. In the ischemic basal ganglia, the peak of VCAM-1 expression occurred in Cerebral ischemia 1h / reperfusion 4h. Infiltration of monocytes / macrophages occurred in ischemic area of cortex at 1 h / reperfusion for 8 h and reached the peak at 16 h after reperfusion; in ischemic basal ganglia, infiltration occurred at 1 h of cerebral ischemia and peak of infiltration In ischemia 1h / reperfusion 8h. CONCLUSION: The expression of vascular endothelial cell adhesion molecules and infiltration of monocytes / macrophages in the basal ganglia are more marked than those in the cortex. Tip basal ganglia brain response to cerebral ischemia than sensitive.
