目的 从人羊水中提取和纯化肺表面活性物质相关蛋白A(SP-A)。方法 收集11例产妇的羊水共4000ml,采用 萃取和透析的方法提纯人羊水中SP-A并测定其生物学活性。结果 获得 SP-A 12 mg(共 20 ml);SDS-PAGE显示,提纯 后的SP-A的相对分子质量为 31 000,只显示1条条带;Western blotting也显示1条特异性反应条带;将其加入人工合 成的磷脂中表现为表面活性明显增加,γmin由7.7±0.3降至1.5±0.22。结论 用萃取和透析方法能较好地提纯SP-A.且纯 度较高,并能较好地保持其生物活性。
Objective To extract and purify pulmonary surfactant associated protein A (SP-A) from human amniotic fluid. Methods A total of 4000 ml of amniotic fluid collected from 11 mothers were collected. The SP-A of human amniotic fluid was purified by extraction and dialysis, and its biological activity was determined. Results The SP-A 12 mg (20 ml total) was obtained. SDS-PAGE showed that the relative molecular mass of the purified SP-A was 31 000, showing only one band. Western blotting also showed one specific band The surface activity of the synthesized phospholipids increased obviously from 7.7 ± 0.3 to 1.5 ± 0.22. Conclusion The extraction and dialysis methods can better purify SP-A. And high purity, and can better maintain its biological activity.
