甲基四氢叶酸-同型半胱氨酸甲基转移酶(B_(12)-甲基转移酶)在 DNA 和蛋白质合成中起着重要作用。此酶在造血细胞中的含量很低,用 Dowex-I-Cl 柱层测定是不易做到的。至今对正常的和白血病造血细胞的B_(12)-甲基转移酶活性测定的报告只有二组材料,且其结果限不一致。近采 Clark 等报告了用溴化氰测定该酶活性的方法。此法持异而简单,尤其对造血细胞酶活性很低的测量更为适宜。它能在几小时内同时分析10个以上的样品,可用于临床及科研工作。通过对该酶活性的测定,可了解造血细胞的维生素 B_(12)及叶酸的代谢情况。
MTHFR-homocysteine methyltransferase (B_(12)-methyltransferase) plays an important role in DNA and protein synthesis. The content of this enzyme in hematopoietic cells is very low and it is not easy to measure with Dowex-I-Cl column. Until now, only two groups of materials have been reported for the determination of B_(12)-methyltransferase activity in normal and leukemic hematopoietic cells, and their results are inconsistent. Kei et al. reported the use of cyanogen bromide to determine the activity of this enzyme. This method is different and simple, especially for the measurement of low activity of hematopoietic cells. It can analyze more than 10 samples simultaneously within a few hours and can be used for clinical and scientific research. By measuring the activity of this enzyme, we can understand the metabolism of vitamin B12 and folic acid in hematopoietic cells.
