目的 研究大鼠肥大细胞表面Fas的表达情况及其功能。方法 应用RT PCR和免疫印迹法检测Fas的转录和表达 ,并用免疫细胞化学法定位 ;应用annexinⅤ流式细胞仪检测经抗大鼠Fas多克隆抗体诱导的RBL 2H3细胞的凋亡情况。结果 RT PCR扩增出Fas细胞外区 4 74bp片段 ,免疫印迹证实肥大细胞有蛋白表达 ,免疫细胞化学证实肥大细胞膜表面有Fas表达 ,annexinⅤ流式细胞仪检测发现在抗大鼠Fas多克隆抗体的诱导下RBL 2H3出现凋亡。结论 大鼠肥大细胞表面表达Fas,通过激活Fas途径可诱导肥大细胞凋亡。
Objective To study the expression of Fas on the surface of mast cells in rats and its function. Methods The transcription and expression of Fas were detected by RT-PCR and Western blotting. The expression of Fas was detected by immunocytochemistry. The apoptosis of RBL 2H3 cells induced by anti-rat Fas polyclonal antibody was detected by annexinⅤ flow cytometry. Results The 4 74bp fragment of Fas extracellular domain was amplified by RT-PCR. The protein expression of mast cells was confirmed by immunoblotting, and the expression of Fas was confirmed by immunocytochemistry. The results of annexinⅤ flow cytometry showed that in anti-rat Fas polyclonal antibody Induction of apoptosis in RBL 2H3. Conclusion The expression of Fas on the surface of mast cells in rats may induce the apoptosis of mast cells by activating the Fas pathway.
