目的观察Vβ13mRNA和Treg细胞在大鼠细小病毒(KRV)联合巨细胞病毒(CMV)感染大鼠的脾淋巴细胞中的表达以及海藻提取物对1型糖尿病的治疗效果。方法 LEW.1WR1大鼠40只,随机分成2组,应用CMV联合KRV建立大鼠1型糖尿病模型。治疗组大鼠每日口服海藻提取物共40d;成模组用等量无菌生理盐水替代同步进行。RT-PCR检测脾脏Vβ13mRNA的表达,流式细胞术检测脾细胞Vβ13~+CD4~+CD25~+Treg细胞亚群比例;HE染色观察糖尿病胰岛的病变情况。结果实验组成模率为90%(18/20),成模血糖平均水平在3.15g/L;实验组Vβ13的表达高于治疗组。实验组中CD4~+CD25~+Treg细胞的比例为6.2%,治疗组为13.5%(P<0.05)。结论感染巨细胞病毒的大鼠Vβ13 mRNA的表达在糖尿病初期显著增高。Vβ13~+CD4~+CD25~+Treg细胞亚群比例较低。海藻提取物提高了LEW.1WR1大鼠Treg细胞的表达,可用于治疗糖尿病。
Objective To observe the expression of Vβ13 mRNA and Treg cells in splenic lymphocytes of rats infected with cytomegalovirus (CMV) and rat parvovirus (CMV) and the therapeutic effect of seaweed extract on type 1 diabetes mellitus. Methods Forty LEW.1WR1 rats were randomly divided into two groups. The rat model of type 1 diabetes was established by CMV combined with KRV. The rats in the treatment group were treated with oral algae extract for 40 days daily, and the rats in the treatment group were treated with the same amount of sterile saline instead. The expression of Vβ13 mRNA in spleen was detected by RT-PCR. The proportion of Vβ13 ~ + CD4 ~ + CD25 ~ + Treg subsets in splenocytes was detected by flow cytometry. The pathological changes of diabetic islets were observed by HE staining. Results The rate of experimental group was 90% (18/20) and the mean blood glucose was 3.15g / L. The expression of Vβ13 in experimental group was higher than that of the treatment group. The proportion of CD4 ~ + CD25 ~ + Treg cells in the experimental group was 6.2%, and the treatment group was 13.5% (P <0.05). Conclusion The expression of Vβ13 mRNA in rats infected with cytomegalovirus was significantly increased in the early stage of diabetes. Vβ13 ~ + CD4 ~ + CD25 ~ + Treg cell subsets lower proportion. Seaweed extract enhances the expression of Treg cells in LEW.1WR1 rat and can be used to treat diabetes mellitus.
