应用植物组织培养技术,探讨利用脱毒种蒜茎尖周围鳞茎组织进行扩繁的新途径。结果表明:以MS为基本培养基,诱导鳞茎愈伤组织形成及分化幼芽的最佳激素浓度组合为2,4—D0.1mg/L+BA0.3mg/L,切割幼芽接种于NAA;BA不同激素浓度处理的培养基中,促使幼苗株高,叶片生长的最佳激素浓度组合为NAA0.05mg/+BA0.5mg/L。因此,切取脱毒种蒜茎尖周围0.2—0.3mm鳞茎组织,经愈伤组织诱导,幼芽分化,幼苗生长三个阶段110天培养,可获得4株试管成苗,并且经幼苗根尖染色体压片,镜检,未发现有任何染色体变异。
