[目的]寻找一条降低橡胶树树根组织培养中外植体污染的有效途径。[方法]对巴西橡胶树树根进行组织培养,取林段断根处理30d后的新根为外植体,采用1g/L多菌灵、0.1%HgCl2和不同浓度益培隆对其进行了消毒试验。[结果]在常规消毒前先采用1g/L多菌灵对外植体处理2.5h,再用75%乙醇消毒30s及0.1%HgCl2消毒6min,然后接种到添加0.1%益培隆的固体愈伤诱导培养基上,培养30d后外植体污染率下降至44.59%,存活率达25.60%。[结论]该研究为降低橡胶树树根组织培养中外植体污染提供了理论依据。
[Objective] The research aimed to find an effective way to reduce the contamination of explants in the root tissue culture of rubber tree. [Method] Tissue culture of the roots of Hevea brasiliensis was carried out. The new roots were taken as explants 30 days after the roots were cut off, and disinfected with 1 g / L carbendazim, 0.1% HgCl2 and different concentrations of iperidone . [Result] The explants were treated with 1 g / L carbendazim for 2.5 h prior to routine disinfection and sterilized with 75% ethanol for 30 s and 0.1% HgCl2 for 6 min, then inoculated into solid callus with 0.1% After 30 days of culture, the explant contamination rate dropped to 44.59% and the survival rate reached 25.60%. [Conclusion] The study provided the theoretical basis for reducing the contamination of explants in root tissue culture of rubber tree.
