目的:研究还原型谷胱甘肽(GSH)对大鼠骨髓基质细胞(BMSCs)胶质纤维酸性蛋白(GFAP)mRNA及蛋白表达的影响。方法:传3代骨髓基质细胞随机分为GSH诱导组和对照组。GSH诱导组加入10 mmol·L~(-1)GSH 20μL,对照组加入生理盐水20μL,两组使用的培养基和处理过程相同。于加入GSH和生理盐水后6、12和24 h观察记录细胞的生存情况及细胞的形态变化。采用实时荧光定量PCR和Western blot法检测各组诱导后细胞GFAP mRNA及蛋白表达水平的变化。结果:经GSH诱导后BMSCs形态发生明显改变,GSH诱导组GFAP mRNA及蛋白表达水平显著高于对照组(P<0.05),以诱导12 h后变化最为明显。结论:BMSCs经GSH诱导后GFAP mRNA及蛋白表达量显著增多。
Objective: To study the effect of reduced glutathione (GSH) on glial fibrillary acidic protein (GFAP) mRNA and protein expression in rat bone marrow stromal cells (BMSCs). Methods: The third passage bone marrow stromal cells were randomly divided into GSH induction group and control group. In the GSH induction group, 20 μL of 10 mmol·L -1 GSH was added, and 20 μL of normal saline was added to the control group. The medium used in the two groups was the same as the treatment. The survival of the recorded cells and the morphological changes of the cells were observed at 6, 12 and 24 h after the addition of GSH and saline. The changes of GFAP mRNA and protein expression in each group were detected by real-time fluorescence quantitative PCR and Western blot. Results: The morphology of BMSCs was significantly changed after GSH induction. The expression of GFAP mRNA and protein in GSH-induced group was significantly higher than that in control group (P <0.05). Conclusion: The expression of GFAP mRNA and protein in BMSCs induced by GSH is significantly increased.
