用CFDA/AM荧光探针负载离体培养的大鼠心肌细胞 ,在激光扫描共聚焦显微镜下观察细胞内胞间通讯的变化。结果发现不同浓度和时间的FGF - 2对原代培养的新生大鼠的心室肌细胞的胞间通讯的影响不同 ,lng/m1组在FGF— 2作用第l天和第 4天对细胞的胞间通讯无明显变化影响 ;10ng/ml和 10 0ng/ml在FGF— 2作用的第 1天的平均荧光恢复速率明显下降 ,在作用后的第 4天 ,平均荧光恢复速率依然显著低于对照组。 10 0 0ng/ml的FGF - 2作用于心肌细胞的第 1天 ,平均荧光恢复速率无显著变化 ,第 4天平均荧光恢复速率则明显增加。
The cultured rat cardiomyocytes were loaded with CFDA / AM fluorescent probe and the changes of intracellular communication were observed under laser scanning confocal microscopy. The results showed that FGF - 2 at different concentration and time had different effects on the intercellular communication of ventricular myocytes in primary cultured neonatal rats. Cells in lng / m1 group were cultured on day 1 and day 4 of FGF - 2 There was no significant change between the two groups. The mean fluorescence recovery rate of 10ng / ml and 100ng / ml on the first day after FGF-2 treatment was significantly decreased. On the fourth day after the treatment, the average fluorescence recovery rate was still significantly lower than that of the control group . On the 1st day of cardiomyocytes treated with FGF - 2 at level of 100ng / ml, the average fluorescence recovery rate did not change significantly, and the average fluorescence recovery rate increased significantly on the 4th day.
